Abstract

Određivanje biogenih amina u vinu važno je zbog toksikoloških implikacija koje mogu biti izazvane njihovom konzumacijom, kao i zbog povezanosti njihove koncentracije u vinu s lošim higijenskim uvjetima tijekom procesa vinifikacije. Među brojnim metodama razvijenim u tu svrhu, najčešće se koriste HPLC metode s derivatizacijom i fluorimetrijskom detekcijom. Cilj ovog rada bila je validacija HPLC metode za određivanje udjela biogenih amina u crnom vinu. Tijekom validacije ispitivani su sljedeći parametri: selektivnost, preciznost, linearnost, granica dokazivanja i granica kvantifikacije. Postupak validacije je pokazao da metoda, iako osjetljiva, zadovoljava postavljene zahtjeve i smatra se prikladnom za predviđenu svrhu.

Abstract (english)

Determination of biogenic amines in wine is of great importance due to the toxicological implications which may be caused by their consumption, as well as their presence in wine being in correlation with poor hygienic conditions during the process of vinification. Among numerous methods developed for this purpose, HPLC methods that include derivatisation and fluorimetric detection are used most commonly. The aim of this study was the validation of HPLC method for determination of biogenic amines content in red wine. During the validation process, the following parameters have been examined: selectivity, precision, linearity, limit of determination and limit of quantification. The process of validation has shown that the method, although refined, complements its fitted requirements, and is considered suitable for its intended purpose.
