Sažetak | Povećana potražnja za 2,3-butandiolom (2,3-BTD) iziskuje nove metode proizvodnje. U ovom radu provedena je biotehnološka proizvodnja 2,3-BTD-a pomoću bakterije Paenibacillus polymyxa DSM 742. Uzgoj u tikvicama proveden je sa i bez dodatka dodatnog izvora dušika. Prinosi u podlozi s glukozom, s glukozom i ksilozom kao izvorima ugljika te u onim pripremljenim od tekućih dijelova hidrolizata konoplje, bukve i jele su redom iznosili 6,1 g/L, 5 g/L, 0,17 g/L, 0,4 g/L i 0,15 g/L, a uz dodatak dodatnog izvora dušika 6,5 g/L, 5,3 g/L, 0,17 g/L, 0,45 g/L i 0,23 g/L. Time je pokazano kako dodatak dodatnog izvora dušika pozitivno utječe na proizvodnju 2,3-BTD-a. U bioreaktoru s čvrstim slojem proveden je uzgoj pri različitim visinama sloja te su prinosi pri visini stupca čvrstog sloja od 7, 10,5 i 13 cm iznosili 0,31 g/L, 0,4 g/L i 0,7 g/L. Zaključno, povećanje visine čvrstog sloja pozitivno utječe na uspješnost bioprocesa. |
Sažetak (engleski) | Increasing demand for 2,3-butanediol (2,3-BTD) requires new methods of it's production. In this research cultivation of bacteria Paenibacillus polymyxa DSM 742 was carried out. Cultivation in flasks was carried out with and without addition of extra nitrogen source. Yields in medium containing just glucose, glucose and xylose and in mediums gained from liquid hydrolyzate of industrial hemp, beech and fir were: 6.1 g/L, 5 g/L, 0.17 g/L, 0.4 g/L and 0.15 g/L, and in those consisting extra nitrogen source 6.5 g/L, 5.3 g/L, 0.17 g/L, 0.45 g/L and 0.23 g/L respectively. In conclusion, addition of extra nitrogen source has positive impact on 2,3-BTD production. Cultivations in fix-bed bioreactor were carried out at a height of solid layer 7 cm, 10.5 cm and 13 cm and therefore yield amounted 0.31 g/L, 0.4 g/L and 0.7 g/L. In conclusion, increase of height of solid layer rises fficacy of bioproces. |
