Abstract

Vilazodone je farmaceutski pripravak s primjenom u liječenju kliničke depresije. Prilikom sinteze Etil 5-(piperazin-1-il) benzofuran-2-karboksilata (PBCE), jednog od ishodišnih spojeva u sintezi Vilazodona zamijećena je prisutnost nepoznatog onečišćenja RRT 1,37. U sklopu ovog diplomskog rada provedena je sinteza, izolacija i karakterizacija prije spomenutog onečišćenja, kao i ispitivanje procesnih parametara na tijek i konačne produkte reakcije. Natrijev karbonat korišten je u reakcijama priprave PBCE kao baza, dok je u reakcijama sinteze onečišćenja kao baza korišten trietilamin. Čistoća izoliranih spojeva određena je HPLC analizom, a na temelju LC-MS analize i NMR spektroskopije potvrđena je struktura onečišćenja.

Abstract (english)

Vilazodone is a pharmaceutical drug used in the treatment of clinical depression. During the synthesis of ethyl 5-(piperazin-1-yl) benzofuran-2-carboxylate (PBCE), a precursor in the synthesis of Vilazodone, an unknown impurity with a relative retention time of 1,37 was detected. In this graduate thesis, synthesis, isolation and characterization of the impurity RRT 1,37 was performed. Furthermore, the assaying of the influence of the reaction process paramteres on the course of the reaction and final reaction products was carried out. Sodium carbonate was used as a deprotonating base in the PBCE synthesis reactions, while the triethylamine was used as a deprotonating base for the synthesis of the impurity RRT 1,37. The purity of the isolated products was confirmed using HPLC method. Spectral analysis of the impurity was carried out using LCMS and NMR spectroscopy.
